Methotrexate is a widely used antimetabolite and antifolate agent indicated for the treatment of various cancers and autoimmune disorders. It inhibits dihydrofolate reductase, thereby interfering with DNA synthesis, repair, and cellular replication.
Methotrexate competitively inhibits the enzyme dihydrofolate reductase (DHFR), leading to reduced formation of tetrahydrofolate. This results in impaired synthesis of thymidylate and purine nucleotides, ultimately inhibiting DNA and RNA synthesis. Rapidly dividing cells, such as cancer cells and activated immune cells, are particularly affected.
