Vinorelbine is derived from the periwinkle plant (Vinca rosea) and is considered a third-generation vinca alkaloid. It was developed to have a more favorable toxicity profile, specifically reduced neurotoxicity, compared to earlier agents like vincristine and vinblastine.
Vinorelbine tartrate primarily exerts its anticancer activity by disrupting the cell cycle during mitosis. It binds to tubulin, the protein subunit responsible for the formation of microtubules, thereby inhibiting the polymerization of tubulin into functional microtubules. Since microtubules are essential for maintaining cell structure and forming the mitotic spindle apparatus, their disruption prevents proper spindle assembly and arrests the cell cycle at metaphase. This mitotic arrest interferes with cell division and ultimately triggers programmed cell death (apoptosis) in rapidly proliferating cancer cells.
